Determination of hexanal and 2-butanone in human serum using magnetic in-tube solid phase micro-extraction coupled with high performance liquid chromatography

Abstract:In this study, C8 functionalized magnetic mesoporous silica spheres (C8-Fe3O4@mSiO2) was prepared and placed in a polytetrafloroethylene tube for magnetic in-tube micro-solid-phase extraction coupled with in-situ derivatization (MIT-MSPE-ISD) for the determination of hexanal and 2-butanone in human serum. Using a magnet to control the adsorbents moving along the inner wall of PTFE tube, the simultaneous extraction and derivation without any other pretreatment was acquired. Under optimized conditions, the limits of detection (LODs) were 1.0 and 1.2 μmol·L-1 for hexanal and 2-butanone, respectively. The relative recoveries ranged from 78.4% to 92.4% with the intra-day and inter-day relative standard deviations (RSDs) less than 7.9%. The proposed method was successfully applied to determine endogenous hexanal and 2-buanone in human serum from healthy persons and liver cancer patients.
