Abstract:Three dimensional structure of DNA in solution has an important significance for understanding the functions of DNA and the misteries of living science.In this paper the methodlogy,strategy and basic principles of structure determination of nuclei acids by 2D NMR techniques have been discussed.From the basic structure of DNA,2D NOE spectroscopy and other NMR methods are applied to the determination of data about interproton distances,bond length and bond angles.Then three dimensional structure of DNA in solution can be obtained by data processing techniques,such as Distance Geometry and Molecular Dynamics.
