Optimization of extraction for composition from Laurocerasus phaeosticta leaf by response surface methodology——total polyphenols, total flavonoids and DPPH radical scavenging activity

Abstract:In order to search for a new natural antioxidant, the total polyphenols, flavonoids and DPPH radical scavenging activity from Laurocerasus phaeosticta leaves were determined based on the single factor experiment method, central composite design and the response surface methodology. The results showed that the optimum extraction conditions was 60% alcohol with extraction temperature of 48℃ and extraction time of 4 d. In the above conditions, the total polyphenol content, total flavonoids content were 404.73 μg GAE g/g DW and 870.37 μg RE/g DW, respectively. DPPH radical scavenging rate was 32.46%. The relative error between the tested value and the predicted value was less than 5%, which indicated that the second-order polynomial model established by the response surface method was effective. Thus, it showed that the ethanolic extraction of Laurocerasus phaeosticta leaves had good antioxidant activity.
