Abstract:Coomassie brilliant bule-G250 is able to react with bovine serum protein and form a blue complex. There was a corresponding relationship between protein concentration and product color depth. On the basis of error transfer law, the phone colorimetric method was used to investigate the external factors influencing sampling such as illumination and phone types. And a new method of protein content determination was established. Results showed that under the optimum conditions the standard deviation XB had a sinificant linear relationship with the concentration of protein at the linear range of 1~7 μg/mL. The relative standard deviation of standard sample determination was 3.47% (n=8), and the limit of detection was 0.67 μg/mL. The standard recovery experiment was conducted on the water-soluble protein of nuts such as peanuts, sunflower seed, pumpkin seed. The recovery was 100.3%~107.5%. This method had the advantages of simplicity, rapidity, high sensitivity and good recovery. and which would be used to determine the content of water-soluble protein in nuts.
