Circulating column chromatographic extraction of total saponin and polysaccharide of Shuangcishen Capsule

Abstract:The process conditions of extracting total saponins and polysaccharides from Shuangcishen Capsules by column chromatography cycle were explored,and the activity of total saponins in vitro were also studied. Based on the extraction rate of total saponins and polysaccharides in the extract, the optimal extraction process was determined by investigating factors such as extraction solvent, suction rate, soaking equilibrium time and elution flow rate. The results showed that the total saponins and polysaccharide extraction solvent were 40% ethanol solution and pure water, respectively. The absorption rates were 3.8 mL·g-1 and 3.6 mL·g-1, respectively,with the soaking equilibrium time of 2 h and the sampling rate of 0.4 mL·min-1. Under these conditions, the total saponins and polysaccharides were continuously extracted in 2 rounds, and the extraction rates were all above 95%. The extracts were mixed and the total saponins and polysaccharides were separated by alcohol precipitation. The contents of total saponins and polysaccharides in the extract were 18.05% and 48.37%, respectively. The crude polysaccharide was purified by two times of water-soluble alcohol precipitation, with the content increasing to 69.91%.The results of the antioxidant test showed that the extract of Shuangcishen had certain antioxidant activity. The total saponins and polysaccharides were extracted by column chromatography cycle combined with low extraction temperature, low ethanol consumption, low energy consumption and simple operation, which offered a foundation for further research of Shuangcishen Capsules.
