Abstract：According to the known sequence of human ob gene cDNA, the gene coding the leptin was synthesized and cloned into pUC18 vector after reasonable primer designing, DNA strain extending, PCR reaction and molecular cloning. Sequence analysis and RE digest results showed the ob gene was synthesized and cloned correctly.
