Purification technology of total flavonods in Rosa davurica Pall. with polyamide resin methods

Abstract:The purification technology of total flavonoids (TFs) in Rosa davurica Pall. with polyamide resin methods by dynamic adsorption-desorption were explored. The purification process of TFs was optimized by single factor and orthogonal test using absorbance, adsorption rate, desorption quantity and desorption rate as indexes for investigation. The results showed that the optimum purification conditions of TFs in Rosa davurica Pall. were as follows: for sample solution, the mass concentration of total flavonoids, loading quantity, pH value and adsorption velocity were 0.9 mg/mL, 19.5 mL/g, 2~4 and 1.5~2.0 mL/min, respectively; 70% (volume fraction) ethanol solution was used as the eluent, of which the desorption velocity, pH value and quantity were 1.0~1.5 mL/min, 8~9 and 12 mL/g, respectively. Under the above purification conditions, the average absorbance of polyamide resin to total flavonoids was 11.159 mg/g, while the average desorption quantity to total flavonoids was 10.204 mg/g. The average desorption rate was 94.44% and the purity of TFs in Rosa davurica Pall. extracts elevated from 26.32% to above 75%. The results provided reference for further development and utilization of Rosa davurica Pall.
