Bioinformafics analysis of human IDE gene promoter region

Abstract:This study was conducted to analyze 5’ regulatory region of human IDE gene by bioinformatics software to obtain the characteristics of the potential promoter, CpG island and transcription factor binding site of human IDE gene. The 5’ regulatory region 2 000 bp sequence of human IDE gene was successfully obtained from UCSC genome database. The number of promoters predicted by software including Promoter 2.0, FPROM and NNPP were 3, 2 and 6, respectively. When relative profile score threshold was 80%, 85%, 90%, 95% and 100%, number of possible transcription factor binding sites in the 5’ regulatory region of human IDE gene which were predicted by JASPAR online software were 5170,1771,454,87 and 5, respectively. When relative profile score threshold was 80%, 5 potential TCF7L2 transcription factor binding sites in the 5’ regulatory region of human IDE were found using JASPAR online software. Using phylogenetic footprinting and LAGAN prediction method, 14 transcription factor binding sites were identified in the conserved region of human and mouse homologous IDE gene promoters, including SPI-1, cap, c-FOS, FREAC-3, c-ETS, Cdxa, HSF2 and et al. A CpG island was found in the upstream of transcription initiation point at 5 ’end of human IDE gene, locating between 1 303-1 705 bp, with a size of 403 bp. The biological information of human IDE gene promoter, CpG island and transcription factor binding site had been obtained, which laid a foundation for further gene expression regulation experiments.
