Effect and mechanism of ethanol extract of Clerodendrum fortunatum L. on asthma-related symptoms

Abstract:Asthma is an inflammatory disease characterized by airway hyperreactivity (airway hyperresponsiveness, AHR), airway remodeling and airway inflammation. Clerodendrum fortunatum L. is one of the ingredients of Xizhuan Pill. In the current study, the role of ethanolic extract of Clerodendrum fortunatum L. (EECF) in the relief of asthma-related symptoms were investigated. The effects of EECF on AHR were investigated by tension testing and respiratory system resistance experiments. The results showed that EECF could reduce intracellular Ca2+ levels by affecting L-VDCCs, NSCCs, NCX ion channels, relax precontracted airway smooth muscle induced by high potassium or ACh, and relieve AHR. In order to further explore the alleviating effects of EECF on airway remodeling and airway inflammation, asthma mice model was established, and a series of experiments were conducted. The results showed that EECF could improve the pulmonary swelling caused by asthma and reduce the respiratory resistance of asthmatic mice. EECF can also reverse Th1/Th2 imbalance, relieve airway remodeling and airway inflammation. The study elucidated the potential therapeutic property of EECF in asthma treatment and the possible underlying mechanism, suggesting the potential application of EECF in the treatment of asthma.
