Abstract:Prevention and treatment for the infection of methicillin-resistant Staphylococcus aureus (MRSA) is a clinical challenge. To fight against the antibiotic resistance, a new medication was studied in this paper, which is the drug combination of a TLR2 agonist Pam3CSK4 and berberine. The combination was able to defend against MRSA through two ways,activation of immunity and antibacterial effect of Chinese traditional medicine. The survival, MRSA clearance and secretion of specific IgG and overall IgM of mice were investigated under MRSA attack when the two drugs were treated alone or together. The secondary proliferation of lymphocytes in vitro was also detected to study the influence of drugs. The results suggested that the combination of Pam3CSK4 and berberine would improve the survival rate of mice under lethal MRSA attack, and enhance the ability of MRSA clearance. Pam3CSK4 was able to suppress the secretion of MRSA-specific IgG in mice, while stimulate the secretion of overall IgM. The secondary proliferation of CD4+ , CD8+ T cells and B cells were also stimulated in vitro. As for berberine, the proliferation of CD4+ T cells and B cells in vitro were promoted.
