Synthesis, structure and bioactivity of 4-{6-amino-2-[(4-cyanophenyl) amino]pyrimidin-4-yl oxy}-3,5-dimethylbenzonitrile and its salts

Abstract:The compound 4-{6-amino-2-[(4-cyanophenyl) amino] pyrimidin-4-yl oxy}-3,5-dimethylbenzonitrile (compound 3) was synthesized with 4,6-rimethylpyrimidine, 3,5-dimethyl-4-hydroxybenzonitrile, p-aminobenzonitrile and ammonia as raw materials, and structurally characterized by 1H NMR and 13C NMR. In order to further study the structure and pharmacological activity, its hydrochloride (compound 4a) and sulfate (compound 4b) were prepared, and their crystal structures were studied by X-ray diffraction single crystal structure analysis. The preliminary biological test shows that the title compound exhibited inhibitory activity on the replication of HIV-1 SF33 virus in vitro.
