Preparation of multi-response carboxymethyl chitosan intelligent hydrogels and their response studies

Abstract:The cross-linking of intelligent hydrogels and their responsiveness have an important effect on the activity of drugs (especially polypeptide or protein drugs) and controlled release. In this paper, mercapto-carboxymethyl chitosan was used to prepare the multi-responsive intelligent hydrogels. The swelling behavior of the hydrogels, the degradation behavior by the colonic enzyme and the reduction behavior by glutathione were studied. The hydrogels showed pH sensitivity, enzyme sensitivity and reduced susceptibility. The thiol content of thiolated carboxymethyl chitosan and the degree of cross-linking of hydogel not only affect the swelling behavior of the hydrogels, but also affect their enzymatic degradation. The reducing sensitivity of the hydrogels depends on their degree of disulfide cross-linking. The hydrogels show the potential for the delivery of polypeptides or protein drugs to colon system.
