Antitumor constituents from roots of Ligularia veitchiana

Abstract:Various chromatographic techniques, including silica gel column chromatography, TLC and HPLC, were used to investigate the chemical constituents of roots of L. veitchiana.Three compounds were isolated and then identified as bakkenolide A (1), phthalic acid isodibutyl ester (2), dibutylphthalate (3), eremophila-7(11),9-dien-8-one (4), 6β,8β-hydroxyeremophil-7(11)-en-12,8α-olide (5), 6β-hydroxyeremophil-7(11)-en-12,8β-olide (6), and 6β-hydroxyeremophil-7(11)-en-12,8α-olide (7) by means of spectroscopic and chemical data. Among them, compounds 1,2,4 and 7 were isolated from this plant for the first time.Compound 1 was the first sesquiterpenoid with a spiro-ring skeleton isolated from L. veitchiana. Compounds 6 and 7 area pair of diastereoisomer. Cytotoxicities of all the compounds against cancer cell lines of HGC-27 were evaluated in vitro by MTT assay, and compound 1 showed a significant growth inhibition of tumor cells HGC-27 with IC50 of 31.10 μg·mL-1.
