Abstract：The Prodenia litura NPV was purified by ultracentrifugation on a discontinuous sucrose gradient. The polyhedral protein was extracted by ultracentrifugation after solubiling with 1/10 0.5 molL~(-1) NaCO-1.4molL~(-1) NaC 1-0.1 molL~(-1)EDTA for two hrs. Two immunoassay methods, couter-immunoelectrophoresis(CIE) and emzyme linked immunosorbent assay(ELISA) were employed to detect the polyhedral protein in definite polyhedra. The results showed that the sensitivety of CIE was 0.025mg PIBs/ml and the sensivety of ELISA was 10ng PIBs/ml.
