Abstract：By PCR, phycoerythrocyanin F gene (pecF) with deletion at N end (pecFn) is amplified from Mastigocladus laminosus DNA, and DNA pecFn is 486 bp. DNA pecFn is the inserted into the multiclonal site of pBluescript to give pBlu pecFn, which is confirmed with sequencing. Then pecFn is transferred to expression vector pGEMD to give pGEMD pecFn, confirmed by physical mapping. Vector pGEMD pecFn is transformed into E.coli BL21(DE3). After induction of IPTG, pecFn was overexpressed to give the protein of 18 kDa in inclusion body.
