Abstract:Notch1 gene of human esophageal squamous cell carcinoma (ESCC) and normal esophageal tissues were amplified by RT-PCR method, respectively. The results showed that Notch1 gene of human ESCC and normal esophageal tissues both could express in transcription level. In addition, the expression of Notch1 protein was detected by immunohistochemical Streptavidin-Biotin Complex (SP) stain in 35 ESCC, 16 paracancerous and 35 normal mucous epithelium tissues. The results revealed that the positive rates of the expression of Notch1 protein in normal esophageal mucous epithelium tissues and precancerous lesions were 82.9% and 68.7%, respectively, which did not differ statistically(P>0.05), however, both of them were significantly higher than 37.1% in ESCC (P<0.05). The low expression of Notch1 protein in ESCC did not correlate with diameter of tumors, tumor stage and the site of tumors (P>0.05), but correlate significantly with lymph node metastasis (P<0.05). It can be concluded that Notch1 protein was over expressed and low expressed in normal esophageal tissues and human ESCC, respectively. This research will lay good foundation for further studying the effect of the expression of notch1 on the development and progression of ESCC.
